NR1D1 and PBP influence survival of ERBB2-positive cells. (A) BT474 cells were transfected with luciferase (control), PBP and NR1D1 shRNAs and co-transfected with GFP. Green cells were counted at intervals and expressed as percentage of control (*P<0.03). (B) BT474 cells were transfected with luciferase (control), PBP or NR1D1 shRNAs for 48h. Immunofluorescence was performed for cleaved Caspase-3 signal; cell nuclei were stained with Hoechst 33243. Cells were counted for cleaved Caspase-3 signal (*P<0.0003). (C) PBP and NR1D1 mRNA levels of BT474 cells transfected with luciferase (control), PBP or NR1D1 shRNAs for 48h, analyzed by qRT-PCRs. (*P<0.003). (D) BT474 cells were transfected with luciferase (control) PBP, NR1D1 or PBP and NR1D1 shRNAs together and co-transfected with GFP. Transfected cells were counted at 24h and 72h and the 72h-to-24h ratio for each shRNA was calculated and expressed as % of the control shRNA (top panel; *P<0.01). Transfected cells were also assayed by immunofluoresence and counted for cleaved Caspase-3 signal at 48h of transfection (bottom panel; *P<0.0001).
