H₂O₂ and IGF-1 affect cell migration in a PML-dependent manner, and PML is a negative regulator of integrin β1 expression. A, MDA-MB-231 cells were treated with H₂O₂ for the times and concentrations indicated. H₂O₂ was then removed and wound-healing assays were performed as described under “Experimental Procedures.” The results shown indicate changes in six independent fields and are representative of at least two independent experiments. B, MDA-MB-231 cells were transfected with control oligonucleotides or a combination of two siRNA against PML. 72 h after transfection cells were treated with 100 μm H₂O₂ for 120 min followed by wound-healing assays as described in A.
