MRG15 is involved in the localization of PALB2 to chromatin. (A,B) U2OS cells transfected with control or the indicated siRNA cocktails were extracted to obtain whole cell lysates (whole), soluble supernatant (sup) and chromatin-enriched pellet (ppt) fractions. Proteins in each fraction were resolved by SDS-PAGE and analyzed by immunoblotting using indicated antibodies. α-tubulin and lamin B are markers for detergent-soluble and insoluble fractions, respectively. Cells were untreated (A) or were harvested 6 hours after exposure to 8 Gy IR (B). (C) Expression of siRNA-resistant MRG15 (F-MRG15*) in U2OS cells to rescue depletion of MRG15 mediated by a siMRG15 cocktail.
