(A,B) U2OS cells transfected with control or the indicated siRNA cocktails were extracted to obtain whole cell lysates (whole), soluble supernatant (sup) and chromatin-enriched pellet (ppt) fractions. Proteins in each fraction were resolved by SDS-PAGE and analyzed by immunoblotting using indicated antibodies. α-tubulin and lamin B are markers for detergent-soluble and insoluble fractions, respectively. Cells were untreated (A) or were harvested 6 hours after exposure to 8 Gy IR (B). (C) Expression of siRNA-resistant MRG15 (F-MRG15*) in U2OS cells to rescue depletion of MRG15 mediated by a siMRG15 cocktail. Cells were separated into whole-cell lysates, soluble supernatants and insoluble fractions and analyzed by immunoblotting for levels of MRG15, PALB2 and BRCA2. α-tubulin and lamin B are markers for detergent-soluble and -insoluble fractions, respectively.
