PB1, LIR, and UBA domains of p62 are vital for ubiquitin and Fau delivery to the mycobacterial phagosomes. (A-D), RAW264.7 cells were transfected with GFP tagged p62 constructs with mutation in PB1, or ΔLIR, or ΔUBA domains. At 24 h after transfection, cells were infected with Alexa-568 labeled BCG and chased for 1 h in complete media. Cells were then subjected to autophagic induction by starvation for 4 h and stained for ubiquitin conjugates (A and C) or Fau (Band D). Data, means ± SEM from at least three independent experiments. For each experiment, at least 50 phagosomes were quantified per condition. **p < 0.01, all relative to starvation control. See also Fig. S6.
