Nematode extract was incubated with dabsyl-N-glycopeptides (A) MM, (B) GnGn, (C) GalGal or (D) EGNEGN (nomenclature based on that of Schachter) in the presence of GDP-Fuc for 5 hours (controls without GDP-Fuc were also performed, data not shown). The MM glycopeptide was apparently not modified, the GnGn substrate is the acceptor for a single fucose residue, the GalGal and βGNβGN for two fucose residues. Laser-induced degradation results, in part, in a decrease of m/z 132 (peaks marked by an asterisk). Hexosaminidase digestion products are indicated with −1HexNAc or −2HexNAc.
