Manganese exposure inhibits IGF-I and IGF-II receptor expression in primary CNS neuron cultures. Rat primary cerebellar neuron cultures were treated with 0, 10, 20, or 40 μM MnCl2 for 48 hours. RNA extracted from the cultured cells was reverse transcribed and used to measure (A) insulin, (C) IGF-II, (B) insulin receptor (R), (C) IGF-I, (D) IGF-IR, and (F) IGF-IIR by qRT-PCR analysis. The mRNA levels were normalized to 18S rRNA measured in the same samples. Graphs depict the mean ± S.E.M. relative levels of gene expression as a function of MnCl2 dose. Trend lines were analyzed using Deming linear regression. Significant P values are indicated in the panels.
