Day 42 Ehd1^-/-testis display a delay in spermatid development and abnormal spermatid clumping. Day 42 testes were Bouin's-fixed, PAS-stained and hematoxylin-counter-stained to visualize the glycoproteins/acrosomes (pink) and nuclei (blue) and analyzed by light microscopy using a 40× objective lens. Stages are labeled with Roman numerals. (AC) WT (+/+) seminiferous tubules displayed evenly spaced round and elongated spermatids. (D) Ehd1^-/-(-/-) seminiferous tubules contained step 9, 10, and 11 elongated spermatids (arrows) and misoriented step 16 elongated spermatids near the basement membrane (circles) and (E) abnormal step 9 spermatids along with aggregates of step 16 spermatids (circles). (F-H) Ehd1^-/-seminiferous tubules displayed membranous wheels or residual bodies (arrows, Rb) containing clumped spermatids near the lumen in stage VIII, (I) misaligned spermatids (circle) and clumped step 16 spermatid nuclei (arrows) in stage X, (J) abnormal step 11 spermatids in stage XI and step 16 spermatids clumping in membranous wheels in stage VIII.
