Comparison of telomere length measurements by TRF Southern blot analysis and telomere PCR. (A) TRF Southern blot analysis of genomic DNA from cultured fibroblasts. The DNA from primary human diploid fibroblast cell lines BJ-hTERT and MRC5 at indicated population-doubling levels (PD) was analyzed by TRF Southern blotting, as described in Methods. Mean telomere TRF lengths in kilobases (Kb) are indicated. (B) Comparison of telomere PCR T/S ratios and TRF measurements for cultured fibroblasts. The MRC5 samples at different PDs yielded a correlation coefficient of 1.0 (solid line; P = 0.0014), and inclusion of the BJ-hTERT sample yielded a correlation coefficient of 0.98 (dashed line; P = 0.0035). (C) Comparisons of telomere PCR T/S ratios and TRF measurements for PBL specimens from AD patients (r = 0.77, P = 0.015). All lines were fit by linear least-squares regression.
