Increase in the Fluorescent Signal Upon Iron Removala
| Construct | % Increase (Iron ➔ Apo)b |
|---|---|
| Fe2hTF | 368c |
| FeNhTF | 74c |
| N-lobe | 266 |
| FeChTF | 71c |
| FeCTEV hTF | 100 |
| C-lobe | 103 |
Iron-containing samples were assayed in 100 mM Hepes, pH 7.4 and apo-samples were generated by incubation in 100 mM Mes buffer, pH 5.6 containing 300 mM KCl and 4 mM EDTA.
Percent increase is calculated as 100 X [(Fapo- Firon)/Firon], in which F is the fluorescence intensity (in counts per second) at the λmax.
