Protein expression levels of human skin fibroblasts following α-irradiation. (A) Western blot analysis of intracellular levels of indicated cytokines 1–4 h after α-irradiation. Actin was used as a protein loading control. Ratios of IL8/Actin and IL33/Actin are indicated. (B) EMSA for determination of nuclear NF-κB DNA-binding activity. Nonspecific IgG or anti-IL33 mAb (2 μg/ml) were introduced into the cell media 1 h before α-irradiation. Nuclear and cytoplasmic proteins were isolated from HSF 2 h after treatment. Nuclear protein was used for EMSA while cytoplasmic protein was used for detection of COX-2 and Actin by Western blot analysis. The p65-p50 NF-κB/NF-Y ratio and the COX-2/Actin ratio are indicated. (C) Western blot analysis of indicated proteins from human skin fibroblasts 16 h after treatment.
