Cerulenin induces changes in tER site morphology and blocks CPY transport from the ER. (A) Wild-type cells expressing Sec13p-GFP (CBY1829) and Sec23p-GFP (CBY2712) were treated with 20 μg/ml cerulenin. (B) Quantification depicting the percentage of cells showing a change in tER site morphology at 1, 2, and 3 h after addition of cerulenin. At least 50 cells were analyzed per condition. (C) Pulse-chase analysis of wild-type cells (CBY740) treated with cerulenin for 2 h. Note that p1 CPY (ER form) was completely converted to the mature form (m) after 20 min of chase in control cells but accumulated in cerulenin-treated cells.
