E protein glycosylation status for DENV-2 wt and loss-of-glycan variants in infected cell lysates. Infected Vero cells and an uninfected control were metabolically labeled from 21 to 24 h p.i., and the E protein was immunoprecipitated from cell lysates using MAb 4G2, followed by treatment with or without PNGaseF and electrophoresis on a 15% SDS-polyacrylamide gel. The DENV-2 E protein in cell lysates is sensitive to partial degradation by proteases, which can give rise to an E protein dimer band in SDS-PAGE profiles.
