Group 3 and group 4 Vif mutants exhibit dominant negative characteristics and interfere with the function of wild-type Vif. The experiment essentially mimicked the one shown in Fig. 2, except that Vif mutants were coexpressed with NL4-3 encoding wild-type Vif. (A, C, E, and G) 293T cells were cotransfected with 3 μg of pNL4-3 wt, 0.5 μg of pcDNA-A3G, and 2.5 μg each of empty vector (NL4-3 wt), Vif wt (wt), or Vif mutants as indicated. Cells transfected with vif-defective NL4-3 and A3G were included as a control [NL4-3vif(−)]. Cell lysates and virus-containing supernatants were processed for immunoblotting as described for Fig. 2.
