HBE1 cells were pretreated with various JAK inhibitors for 3 hours (JAKi1, 1 μM; Jak2i, 50 μM; Jak3i, 80 μM), followed by IFNγ (100 ng/mL) treatment for two hours. Cells were harvested for RNA extraction 24 hours after IFN-γ treatment. rt-QPCR analyses of mRNA expression are presented as fold-induction of DUOX2 (A) or CXCL10 (B) normalized to β-actin by defining 2−ΔΔCt value from the DMSO-treated sample as 1. Data were pooled from five separate experiments and represent mean ± SD. *; p < 0.05
