AR inhibition prevents HG- and/or TNF-α-induced synthesis phase in VSMC. Growth-arrested VSMC were preincubated with 10 μm sorbinil or carrier for 24 h in the presence or absence of HG followed by stimulation with TNF-α (2 nm) for another 24 h. The cells were trypsinized and washed, and DNA was stained with propidium iodide as described in Materials and Methods. Cell cycle analysis was performed with a minimum of 10,000 events per analysis by using FACScan flow cytometer.
