Effect of ICI 118551 and Ad5.CMV.PKIα on salmeterol-enhanced IL-6 and IL-8 release. (A) BEAS-2B cells were either left untreated or were incubated with ICI 118551 (2 µM) for 30 min. Cells were then either left untreated or were incubated with salmeterol (0.1 µM) for a further 1 h prior to either being not stimulated (NS) or stimulated with IL-1β (1 ng·mL−1), or IL-1β (1 ng·mL−1) plus histamine (100 µM) (I + H). (B) BEAS-2B cells were either left uninfected or were infected with an MOI of 100 of Ad5.CMV.PKIα or Ad5.CMV.Null for 24 h. After incubation in serum-free media overnight, cells were then left untreated or were incubated with salmeterol (0.1 µM) for 1 h prior to either being not stimulated (NS) or stimulated with IL-1β (1 ng·mL−1), or IL-1β (1 ng·mL−1) plus histamine (100 µM) (I + H). Cells were harvested after 6 h, and the supernatants were analysed by elisa for IL-6 and IL-8 release.
