Subcellular localization of TprA protein. (A) Cytoplasm-periplasm, total membrane, inner membrane, and outer membrane fractions. Cells of strains W83 (wild type) and KDP159 (tprA::ermF) were fractionated and subjected to SDS-PAGE and immunoblot analysis using anti-TprA antiserum. Lanes: 1, cytoplasm-periplasm fraction; 2, total membrane fraction; 3, inner membrane fraction; 4, outer membrane fraction. (B) Spheroplast and periplasm fractions. Preparation of spheroplasts of P. gingivalis cells was performed as described in Materials and Methods, and the spheroplasts were subjected to proteinase K treatment (Pro K) in the presence or absence of 2% Triton X-100 (TX-100).
