(A) Dependence of yeast sensitivity to BiQs on NDE1 in glucose and glycerol containing media. Spot assays of wild-type, nde1nde2Δ, nde1Δ, nde2Δ and wild type yeast overexpressing NDE1 driven by a GPD promoter (PGPD-NDE1) were spotted in five fold serial dilutions on plates containing glycerol or glucose and either DMSO or BiQ7, BiQ11 or BiQ3 at their IC50 concentrations. Spot assays were performed in duplicate in two separate experiments with representative plates shown. (B) Decreased generation of ROS in the nde1Δnde2Δ mutant following exposure to 5 µM BiQ7. (C) Increased generation of ROS in a yeast overexpressing NDE1 (PGPD-NDE1) following exposure to 1 µM BiQ7. Yeast strains were treated with binaphthoquinones or DMSO for 2h and incubated with DHR 123. The experiment was performed three times with similar results.
