(A) ER chaperones GRP78, CNX and CRT but not PDI were upregulated in Grp94−/− ESCs. Grp94+/+ and −/− ESCs were treated with 300 nM ER stress inducer thapsigargin (Tg) for the indicated time period (hrs). The whole cell lysates were subjected to Western blot. (B) Summary of the Western blot results of panel (A), which was repeated 2 to 3 times. Quantitation of the Western blots was performed using Quantity One software with the level of each chaperone in Grp94+/+ ESCs at 0 hr set as 1.
