Rate of dissociation of S1 from actin-tropomyosin-troponin as a function of ATP concentration at three temperatures. Light scattering was used to measure dissociation of both acrylodan-labeled (open symbols) and pyrene-labeled tropomyosin (solid symbols). Reactions were run at 3°C (diamonds), 5.5°C (triangles up), and 9.9°C (triangles down). Solid lines are hyperbolic fits to acrylodan-tropomyosin at 3°C (bottom line) and to pyrene-labeled tropomyosin at 9.9°C (upper line). Inset: Plot of light-scattering intensity against time, showing dissociation of S1 at 3°C and 2 mM ATP. The heavy solid line is an exponential fit to the part of the curve past the mixing time (∼1 ms). Each tick on the abscissa is 4 ms. Conditions: 4 μM S1, 4 μM actin, 0.86 μM tropomyosin, and 0.86 μM troponin in 20 mM MOPS pH 7, 158 mM KCl, 2 mM EGTA, 4 mM MgCl2, and 1 mM dithiothreitol.
