All experiments were performed in triplicate. B–G, Robustness of response to rapamycin administration parallels the intensity of P-mTOR (C, F) and P-S6 (D, G) expression in both T24 and UMUC3 cells. Western blot analysis of T24 cells (H) and UMUC3 cells (I) after treatment with DMSO (Ctrl) or rapamycin (1 pM to 1 μmol/L) shows a reduction in P-S6K and P-S6 levels at low-dose rapamycin that parallels reduction in cell proliferation.
