In good agreement with the levels of cross-contaminations measured using Western blot (WB) experiments (see supplemental Data 1), envelope membranes appear to contain up to 10% stroma proteins and 6% thylakoid membranes proteins. Contamination thresholds were selected accordingly to evaluate subplastidial localization of the proteins identified within the purified envelope fraction (see supplemental Data 10).
