TrwBΔN70 binds to G4 DNA structures with high affinity. Gel mobility shift assays of different TrwBΔN70-DNA complexes. Reaction mixtures contained 10 fmol of 32P-labeled G4 DNA, ssDNA, dsDNA, or forked DNA substrates in panels A, B, C, or D, respectively. The DNA substrates were incubated with TrwBΔN70 protein at 1, 2, 4, 10, 50, and 100 nm, as described under “Experimental Procedures.” DNA substrates used as molecular markers (MM) are drawn on the left of the gels and correspond to oligonucleotide A (45-mer), a 45-bp-long dsDNA formed by the annealing of nucleotides A and G, a 50-bp-long forked DNA formed by the annealing of nucleotides E and F, and a G4 DNA formed after the appropriate treatment of oligonucleotide B.
