CTRL- and NOX2-RNAi-transfected A549 were subjected to mock or poly I:C transfection for the indicated times. (A) WCE were resolved by SDS-PAGE. Efficiency of NOX2 depletion by NOX2-RNAi was evaluated by immunoblot (IB) using tubulin detection as a loading control. On a separate gel, WCE were analyzed by IB using anti-IRF-3-P-Ser396, anti-IRF-3 and anti-actin antibodies. (B) WCE were resolved by native gel electrophoresis and immunoblotted with anti-IRF-3 antibodies. Representative immunoblots of three different experiments are shown. hpi: hours post infection.
