Analyses of endocytosis. (a) Time-course of FM4-64 internalization via the endocytic pathway. Living wild-type (WT), sst4/vps27Δ (sst4Δ) or sst6Δ cells were labelled with 16 μM FM4-64 on ice for 30 min and then incubated at 30 °C for the indicated periods. (b) Accumulation of Lucifer Yellow CH. Wild-type (WT), sst4/vps27Δ (sst4Δ) or sst6Δ cells were incubated for 60 min at 30 °C in YES medium containing 5 mg ml⁻¹ Lucifer Yellow CH (LY). Cells were then washed and observed by fluorescence microscopy.
