CD2AP associates specifically to the C terminus of Rac1. A–C, peptides encoding the C-terminal hypervariable domain of different Rho family members were used to pull down proteins from cell lysates of Jurkat T-cells (A), primary human umbilical vein endothelial cell (HUVEC) (B), or CD34+ cells (C). Long and short exposed (Exp.) blots are shown. D and E, to test for association of CD2AP to full-length Rac1, bacterially isolated GST-Rac1, a mutant of Rac1 lacking the C terminus (GST-Rac1-ΔC) and GST as a control, were used to pull down proteins from lysates of HeLa cells transfected with CD2AP-GFP (D), or from Jurkat T-cell lysates (E).
