Shear stress-increased SIRT1 is AMPK independent. (A and B) HUVECs were transfected with AMPK α1 and α2 siRNAs (10 nM each), SIRT1 siRNA (20 nM), or with control RNA (20 nM) before being subjected to laminar flow for 16 h. (C and D) MEFs ablated with AMPKα or SIRT1 (i.e., AMPKα^−/− and SIRT1^−/−) were subjected to laminar flow, and cell extracts were collected at the indicated times. (E) Wild-type MEFs were infected with Ad-AMPK-CA [100 multiplicities of infection (MOIs)] or Ad-null for 48 h. Total cell proteins were then resolved by SDS/PAGE and subjected to Western blot analysis with various antibodies as indicated.
