Analysis of VEGF gene expression in TGF-β1-treated HSCs. (a) HSCs were cultured for 24 h in a serum-free medium and were then stimulated with TGF-β1 for 12 h at indicated concentrations. Total RNA was isolated from HSCs using an RNAeasy Mini kit, and VEGF gene expression was measured by real-time PCR. TGF-β1 significantly induced VEGF mRNA expression. VEGF values were normalized to 18S mRNA values. Data are expressed as mean±s.e., *P<0.05. (b) Effect of PTK/ZK on VEGF mRNA induced by TGF-β1. PTK/ZK at 5 or 10 μM concentration was added 4 h before TGF-β1 (1 ng/ml). The VEGF gene expression level was significantly suppressed by PTK/ZK. Data are presented as mean±s.e., *P<0.05.
