PDGF-C induces glomerular endothelial cell proliferation but does not affect mesangial cell proliferation or activation. PDGF-C infusion augmented proliferation of glomerular endothelial cells but not of non-endothelial cells as assessed by counting mitotic figures (A). Numbers of JG12 (brown)/PCNA (blue) double-positive-stained glomerular cells in the PDGF-C-treated group increased in comparison with rats receiving PBS only (B and F, P = 0.055). Proliferating mesangial cells were identified by PCNA (brown)/α-SMA (blue) double staining (C and E) and showed no differences between the groups. Glomerular α-SMA expression, a marker of activated mesangial cells, was not affected by PDGF-C treatment (D and G). Arrows point to double-positive cells.
