Atg9 trafficking is impaired the sec7^ts mutant. (A) WT (FRY162), atg1^ts (FRY170 transformed with the plasmid carrying the carrying the ATG1^ts allele), sec7^ts (FRY326) and sec7^tsatg1^ts (AVY065 transformed with the plasmid carrying the carrying the ATG1^ts allele) cells expressing Atg9-GFP under the control of its authentic promoters were grown at 24°C to an early log phase before being transferred at 37°C for 1 h. Cells were imaged before and after the temperature shift. (B) The experiment described in A was repeated in presence of rapamycin to study Atg9 trafficking upon autophagy activation.
