FcRL3 expression was analyzed by flow cytometry as in Figure 1 in 29 healthy donors and the % FcRL3+ (A) in addition to the FcRL3 MFI (B) within Treg was determined. The FcRL3 MFI was calculated as [FcRL3 MFI – control stain MFI]. PBMC from the same donors were screened for the −169 C/T FCRL3 SNP genotype by a PCR allelic discrimination assay. Donors are grouped according to genotype and shown in the category scatter plots against the % FcRL3+ (A) or FcRL3 MFI (B) within Treg; horizontal bars show the mean of each group. Statistical significance was determined using unpaired Student’s t-test; * indicates that differences were significant (P < 0.05) and *** that differences were extremely significant (P < 0.001).
