Proteins from murine RAW 264.7 macrophages naturally expressing wild-type P2X7 as well as human HEK293 and monkey COS7 cells transfected with human wild-type P2X7 were denatured and treated with either EndoH or PNGase F for 1 h at 37°C as described under Materials and Methods. The proteins were then resolved on polyacrylamide gels containing SDS and immunoblotted for P2X7. Cleavage of EGFR was used as a positive control to show EndoH and PNGase F were active. These data are representative of three or more experiments.
