Results of an anchorage-independent soft agar assay showed that CTD MSK1 (residues 414-738) increased EGF- (A) or TPA (B)-induced neoplastic transformation of JB6 cells in a manner similar to the wildtype MSK1 protein (FL). In contrast, the “full length” CTD MSK1 (residues 414-802) exhibited decreased cell transformation activity. The agar plates were photographed (left panels) and the average colony numbers were calculated (right panels) from 3 separate plates. Data are expressed as means ± S.D. and the asterisk indicates a significant decrease in colony formation (p < 0.05).
