TLR4 induces OPN expression in a PI3K-dependent manner. RAW264.7 macrophages were pretreated with DMSO, 30 μm LY294002, or 100 nm Wortmannin for 40 min and then stimulated with 100 ng/ml LPS for the indicated time periods. Expression level of OPN mRNA was examined by both RT-PCR (A) and quantitative PCR (B). The production of intracellular OPN was detected by Western blot with R&D OPN Ab (C).
