Figure 1. Inhibition of protein synthesis dissociates the SMN complex and impairs its activity.

(A) Immunofluorescence microscopic analysis of SMN complex components was performed before (Control) and after protein synthesis inhibition (CHX) (see also Figure S1 and S2).

(B) Immunofluorescence microscopic analysis as in (A) of the Gemin5-containing subunit.

(C) Extracts were prepared from cells treated with cycloheximide (CHX) or anisomycin (ANS) and analyzed by Western blot using antibodies specific to SMN complex proteins.

(D) Total cell extracts prepared as in (C) were used to measure the activity of the SMN complex. This activity represents the amount of Sm cores formed on biotinylated U4 snRNA. Error bars represent the mean of 2 independent experiments, each performed in triplicate.
