Acid and bile salts increase CDX2 promoter activity by activating NF-κB in NES-B3T and NES-B10T cells. (5A &5B) NF-κB inhibition by Bay 11-705 and site-directed mutagenesis of the NF-κB binding site within the reporter construct pGL-3-CDX2 prevents activation of the CDX2 promoter in both cell lines. (5C &5D) NF-κB inhibition by infection with the adenoviral construct Ad5IκBα-SR prevents activation of the CDX2 promoter; the corresponding empty adenoviral vector served as a control.
