Taurocholate rapidly downregulates PEPCK and G-6-Pase mRNA in the chronic bile fistula rat. Pieces of frozen liver (100 mg) from the chronic bile fistula rats, harvested at various time points (0–4 h, see Fig. 1), were used to extract total mRNA. This mRNA was used for reverse transcription and then for real-time quantitative RT-PCR to quantitate PEPCK and G-6-Pase mRNA as described in “Materials and Methods.“ N = 3 for all points.
