Zinc-mediated disruption of glutathione homeostasis is mediated by the inhibition of glutathione reductase activity. PAECs exposed to ZnCl2 (0–1 mM) were analyzed to determine GSH-to-GSSG ratio (a) and GR enzymatic activity (b). ZnCl2 causes a dose dependent decrease in the GSH-to-GSSG ratio that is correlated with a decrease in GR activity. Confirming the ZnCl2 mediated inhibition of GR, purified bovine GR was inhibited in vitro when exposed to ZnCl2 (0–100 µM, c).
