HeLa cells were infected for 16 h with GFP-expressing Salmonella strains, fixed and immunostained for FK2, P62 or LC3 and LAMP1 as a SCV membrane marker. (A) The percentages of FK2-, P62- or LC3- positive vacuoles were scored. Deletion of sopD2 increased the percentages of vacuoles positives for these markers. These phenotypes were complemented by expression of sopD2 from a plasmid. Values are means ± SD of 3 independent experiments. (B) HeLa cells were imaged by confocal microscopy for GFP (green), LAMP1 (red) and P62 (blue). Scale bar, 5 µm.
