(A) SopD2 blocks the formation of PipB2- and kinesin-1-positive vesicles. HeLa cells were infected for 16 hours with strains over-expressing PipB2-2HA from a plasmid. Fixed cells were immunostained and imaged for HA (green), kinesin heavy chain (red) and GFP (blue). Solid and dotted lines (central rows) delineate cells and nuclei, respectively. PipB2- and kinesin-positive vesicles accumulate at the periphery of cells infected with wild-type and sifA sopD2 strains while these molecules accumulated on sifA and sifA sseJ SCVs. (B) SopD2 negatively regulates the formation of PipB2-positive but LAMP1-negative tubules. HeLa cells were infected with strains (blue) expressing 2HA-tagged PipB2 from the chromosome. Fixed cells were immunostained and imaged for HA (green), LAMP1 (red) and GFP (blue). PipB2- and LAMP1-positive tubule (arrowheads) and PipB2-postive but LAMP1-negative tubule (arrows) are indicated.
