Continuous high frequency stimulation (HFS) of the subthalamic nucleus (STN) evoked a transient release of dopamine in the dorsomedial striatum that peaked within 20 applied pulses at 50 Hz (A, gray line), as compared to an 11-fold greater and more sustained dopamine release to stimulation of dopaminergic axons dorsal to the STN (A, black line; note the right y-axis is 10 × greater than the left y-axis). Despite a 50% (B) to 70% (C) loss in striatal dopamine, HFS of the substantia nigra (SN) or STN (black and gray lines, respectively) evoked reliable and quantifiable release of striatal dopamine in an awake parkinsonian rat model.
