Effects of in vivo MafG and c-Maf knockdown in BDL mice on gene expression, ARE nuclear binding and GSH levels. Mice were subjected to BDL or sham surgery and treated with lentivirus carrying scrambled (SC), MafG, c-Maf or both MafG and c-Maf siRNAs as described in Methods and sacrificed on day 14. Parts A and B show Western blot analyses of MafG, c-Maf, Nrf2, GCLC, GCLM and GS. Part C shows EMSA and supershift analysis for Nrf2 binding to ARE using nuclear proteins from BDL mice treated with SC, MafG, c-Maf or both siRNAs. Part D summarizes hepatic GSH levels in these mice.
