Iron (II) blocks HIF-1 activation by 1 under normoxic conditions. Reporter assay data showing that addition of iron(II) inhibits HIF-1 activation by 1 and by the iron chelator DFO in a dose dependent manner (A). Averages from one representative experiment performed in quadruplicate are shown and the bars represent standard deviation. An asterisk (*) indicates p <0.05 when compared to the control. HIF-1α and HIF-1β proteins in the nuclear extract samples were detected by Western blot (B). These extracts were prepared from T47D cells treated with 1, FeSO4, or both, at the indicated concentrations for 4 h.
