TLC analysis of enzymatic synthesis of UDP-N-acetylglucosamine (UDP-GlcNAc) and UDP-N-acetylgalactosamine (UDP-GalNAc) using N-acetylglucosamine uridyltransferase (GlmU). The enzymatic reactions were carried out for 12 h. The developing solvent used was n-butanol:acetic acid: water = 2:1:1 (v/v/v). The plate was developed twice and stained with p-anisaldehyde sugar stain. Lane: 1 and 5, UDP-GlcNAc; 2 and 6, UTP; 3, N-acetylglucosamine 1-phosphate (GlcNAc-1-P); 4, reaction mixture of GlcNAc-1-P using GlmU; 7, N-acetylgalactosamine 1-phosphate (GalNAc-1-P); and 8, reaction mixture of GalNAc-1-P using GlmU.
