Activation of ROS and LPO in proton-irradiated LE cells. A, equal numbers of rat LE cells were seeded in 96-well plates and cultured for 24 h. Cells were then incubated with 10 μm H2DCF-DA for 3 h in Hanks' balanced salt solution and exposed to different doses of protons (0.1, 1, 2 and 4 Gy), and ROS dichlorofluorescein (DCF) fluorescence was measured. The 2.5 h of post-irradiated values are expressed as dichlorofluorescein fluorescence units. B, LE cells were equally seeded, grown for 24 h, and irradiated with protons (0.1, 1, 2, and 4 Gy), and proteins were extracted at different time points as shown. Fifty microgram of protein from each dose and time point was used to measure LPO levels. Values are means ± S.D. of three experiments performed independently.
