Treatment of human aortic endothelial cells with IL-4 stimulates monocyte migration. Conditioned media (CM) were collected 16 h after treatment of HAEC with either PBS (control) or 10 ng/ml of IL-4. CM from IL-4-treated HAEC were pre-incubated with 10 µg/ml of non-immune isotype control IgG2B (negative control) (IL-4 + IgG) or 10 µg/ml of neutralizing anti-MCP-1 antibody (IL-4 + MCP-1 Ab) for 1 h at 37°C. Chemotactic activities of CM were measured by migration of THP-1 cells using the Chemicon 96-Well Cell Migration Assay Kit.
