Drug inhibition of DNA-PKCS activity. Transformed normal and ATM−/− cells were treated with 10 μM NU7441, a specific inhibitor of DNA-PKCS activity, for 24 h before infection with the four reporter vectors. In negative-control cells, DMSO was added at the same final concentration. Transduction was assayed at 24 h postinfection. Mean decreases for transduction in untreated and NU7441-treated cells for each vector are all significantly different (P = 0.0002 for scAAV-GFP in ATM−/− cells; P < 0.0001 for all the others). Error bars indicate standard deviations.
