Effect of blocking chemokine receptors on eosinophil migration in response to S. stercoralis soluble extract. Eosinophils (1 × 106) were added with IL-5 (30 ng/ml) to the upper chambers of a transwell plate. Chemotaxis was stimulated for 2 h with either eotaxin/CCL11 (80 ng/ml), SDF-1α/CXCL12 (400 μg/ml), MIP–2/CXCL2 (100 ng/ml) or S. stercoralis extract (1 mg/ml) in the lower wells. a Treatment with the CCR3 antagonist, SB328437. b Treatment with the CXCR4 antagonist, AMD3100. c Treatment with the CXCR2 antagonist, SB225002. Data are presented as percentage of migration in relation to untreated control, represented as 100%. These data represent the mean percentage over 5–6 experiments ± standard error of the mean. Numbers above control bars indicate the raw number of untreated migrated cells (arithmetic mean ± standard error of the mean ×103 migrated cells). * p<0.05.
