Efficacy of wild-type and mutant CocE in vivo. (A) CocE protects against cocaine-induced lethality in a time-dependent manner. Wild-type (wt-CocE) or mutant CocE (as indicated) (0.1 mg, 0.3 mg, or 1 mg i.v.) was administered at different time points before cocaine administration (180 mg/kg, i.p.). Each data point represents the percentage of mice (n = 6 for each dosing condition) exhibiting cocaine-induced lethality. (B) Estimated duration of protection for 50% lethality. Time required to reach 50% lethality for CocE and each mutant was measured from the previous panel and was plotted against dosage.
