Monitoring the formation of stable T7 RNAP ECs. Transcription from templates allowed to incubate with T7 RNAP for a defined period of time is proportional to the number of T7 RNAPs successfully loaded onto the templates and awaiting the addition of UTP. The rates at which ECs are forming on ^OT (blue squares) and ^+UT (black circles) do not seem to be significantly different, but ECs form more quickly onto ^UT (red triangles). EC formation occurs more quickly on all three minicircle templates than on a supercoiled, 2536 bp plasmid (green diamonds).
